Transforming growth factor beta-treated human placenta-derived mesenchymal stem cells (TGFβ-hPMSCs) is an experimental cell therapy approach currently in the preclinical research stage. This modality involves the ex vivo treatment of human placental mesenchymal stem cells (hPMSCs) with the cytokine transforming growth factor beta (TGFβ), typically at a concentration of 20 ng/mL, to enhance their therapeutic potential. In preclinical models of thyroid-associated ophthalmopathy (TAO), TGFβ-hPMSCs have demonstrated superior anti-adipogenic, anti-lipogenic, and anti-fibrotic properties compared to untreated hPMSCs. The mechanism of action is mediated through the SMAD 2/3 signaling pathways and the inhibition of SREBP2-HMGCR signaling, which collectively suppress the differentiation of orbital fibroblasts into mature adipocytes and reduce inflammation.
