Transgenic lymphocyte immunization vaccine is an investigational cancer vaccine approach in which genetically modified (transgenic) lymphocytes are infused into a patient to serve as antigen-presenting cells (APCs). These engineered cells synthesize and present specific antigens—such as telomerase or tumor-associated proteins—to the immune system, thereby stimulating an immune response that targets cancer cells expressing those antigens. The mechanism involves both direct presentation by the infused transgenic B or T lymphocytes and cross-presentation by host dendritic cells. This strategy aims to induce robust CD4+ and CD8+ T cell responses against tumors. Preclinical studies have demonstrated its ability to activate cytotoxic T cell responses and provide protection in models of viral infection and cancer. Early-phase clinical trials have explored its use for cancers such as neuroblastoma, showing induction of antitumor immunity with manageable toxicity[1][2][3][4].
