Transition Bio's TOPBP1 program is a preclinical oncology initiative focused on developing small molecule inhibitors that target the DNA topoisomerase II-binding protein 1 (TOPBP1). TOPBP1 is a multi-functional scaffold protein that plays a pivotal role in the DNA damage response (DDR), particularly in the activation of the ATR kinase at stalled replication forks. Transition Bio employs its proprietary "Condensomics" platform—which integrates droplet microfluidics, high-throughput screening, and machine learning—to identify compounds that disrupt the formation or stability of TOPBP1-containing biomolecular condensates. By interfering with these condensates, the therapy aims to compromise the DDR pathway, leading to selective cell death in cancers characterized by genomic instability or specific DNA repair deficiencies, such as ovarian cancer and other solid tumors.
