TranspoCART19 is an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target the CD19 antigen on B-cell malignancies. Distinguishing it from traditional CAR-T therapies that utilize viral vectors (such as lentivirus or retrovirus) for genetic modification, TranspoCART19 is produced using the non-viral **Sleeping Beauty transposon** delivery system. This platform is intended to be a more cost-effective and efficient alternative for generating genetically engineered T cells under GMP conditions. The CAR construct features a **4-1BB** costimulatory domain to enhance T-cell persistence and is co-expressed with a truncated version of the human epidermal growth factor receptor (**hEGFRt**). The hEGFRt serves as both a reporter gene for monitoring and a safety switch, allowing for the targeted elimination of the CAR-T cells through the administration of cetuximab if severe toxicity occurs.
