TrasGEX is a second-generation, glyco-optimized monoclonal antibody derived from trastuzumab. It is engineered to enhance antibody-dependent cellular cytotoxicity (ADCC) while fully retaining the antigen-binding properties of trastuzumab to human epidermal growth factor receptor 2 (HER2). The optimization involves modification of N-glycans with low fucose content, resulting in higher binding affinity to FcγRIIIa on natural killer cells and improved ADCC activity compared to conventional trastuzumab. TrasGEX has demonstrated antitumor activity in patients with advanced HER2-positive carcinomas and may benefit a broader patient population due to its enhanced efficacy across different FcγRIIIa allotypes. The drug was developed using the GlycoExpress platform by Glycotope and has been evaluated in phase I clinical trials for safety, pharmacokinetics, and preliminary efficacy[1][3][5][6][7].
