Overview

Trastuzumab is a humanized monoclonal antibody that targets the extracellular domain of the human epidermal growth factor receptor 2 (HER2). It is primarily used to treat HER2-positive breast cancer and also approved for HER2-positive gastric and gastroesophageal junction cancers. Trastuzumab binds with high affinity to HER2 on tumor cells that overexpress this protein (about 20–30% of breast cancers), inhibiting cell proliferation and survival. Its mechanisms include direct inhibition of HER2-mediated signaling pathways and mediation of antibody-dependent cellular cytotoxicity. The drug was originally developed by Genentech/Roche[1][3][7].
