Overview

This investigational cellular immunotherapy consists of activated and expanded autologous natural killer (NK) cells administered in combination with the HER2-targeted monoclonal antibody trastuzumab. Developed by Xuzhou Medical University, the therapy involves collecting a patient's own NK cells via apheresis and expanding them ex vivo using a K562 feeder cell line genetically modified to express membrane-bound interleukin-15 (IL-15) and 4-1BB ligand (4-1BBL). The treatment regimen typically includes trastuzumab to target HER2-positive cancer cells and subcutaneous interleukin-2 (IL-2) to support NK cell survival and activity. The primary mechanism of action is the enhancement of antibody-dependent cellular cytotoxicity (ADCC), where the expanded NK cells recognize and destroy trastuzumab-coated tumor cells, specifically for the treatment of HER2-positive advanced gastric cancer.
