Trastuzumab + CEF is a combination regimen used primarily in the treatment of HER2-positive breast cancer. Trastuzumab is a recombinant humanized monoclonal antibody that selectively binds to the extracellular domain of the human epidermal growth factor receptor 2 (HER2), inhibiting proliferation and survival of cancer cells that overexpress this protein. The CEF component refers to a chemotherapy regimen consisting of cyclophosphamide, epirubicin, and 5-fluorouracil. This combination exploits both targeted therapy (trastuzumab) and cytotoxic chemotherapy (CEF) to improve outcomes in early-stage and locally advanced HER2-positive breast cancer[1][3][5]. Trastuzumab’s mechanism includes blocking HER2 signaling pathways, inducing antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), internalization/degradation of HER2 receptors, inhibition of DNA repair mechanisms, and suppression of cell cycle progression[1][3][5]. The addition of trastuzumab to standard chemotherapy regimens like CEF has been shown to significantly improve disease-free survival in patients with HER2-positive tumors.
