Trastuzumab + docetaxel is a combination therapy used primarily for the treatment of HER2-positive breast cancer. Trastuzumab is a humanized monoclonal antibody that targets the extracellular domain of the human epidermal growth factor receptor 2 (HER2), which is overexpressed in approximately 20% of breast cancers. By binding to HER2, trastuzumab inhibits downstream signaling pathways such as MAPK and PI3K/Akt, induces cell cycle arrest, and promotes antibody-dependent cellular cytotoxicity (ADCC) against tumor cells[5][6][8]. Docetaxel is a taxoid antineoplastic agent that stabilizes microtubules by binding to β-tubulin, preventing their depolymerization and thereby inhibiting mitosis and cell division[3]. The combination leverages both targeted therapy (trastuzumab) and cytotoxic chemotherapy (docetaxel) for synergistic anti-tumor effects in patients with HER2-positive metastatic or early-stage breast cancer[5][8].
