Drug intelligence / Profile preview
Hanquyou + docetaxel is a combination therapy consisting of Hanquyou (trastuzumab biosimilar, HLX02), a recombinant humanized IgG1 monoclonal antibody targeting human epidermal growth factor receptor 2 (HER2), and docetaxel, a microtubule-stabilizing taxane chemotherapy agent. Developed by Shanghai Henlius Biotech, this combination is used for the treatment of HER2-positive breast cancer, particularly recurrent or metastatic breast cancer. Trastuzumab binds to subdomain IV of the extracellular domain of HER2, inhibiting ligand-independent HER2 homodimerization and heterodimerization, suppressing downstream PI3K/Akt and MAPK signaling pathways, and mediating antibody-dependent cellular cytotoxicity (ADCC). Docetaxel binds to and stabilizes beta-tubulin, preventing microtubule depolymerization, which leads to cell cycle arrest in the G2/M phase and induces apoptosis. Together, the two agents exhibit synergistic antitumor activity by simultaneously blocking HER2-mediated survival pathways and disrupting mitotic spindle assembly.
