A two-drug antineoplastic regimen combining the anti-HER2 monoclonal antibody trastuzumab with the topoisomerase II inhibitor etoposide. Trastuzumab targets ERBB2/HER2 to inhibit oncogenic signaling and mediate immune effector functions, while etoposide induces DNA double-strand breaks by stabilizing the topoisomerase II–DNA cleavable complex. The combination has been explored particularly in HER2-positive metastatic breast cancer, including oral etoposide schedules in heavily pretreated patients, and in ongoing clinical studies in combination backbones for HER2-positive disease.
