HL02 + WBP257 is a combination therapy consisting of two biosimilar monoclonal antibodies developed by Hualan Genetic Engineering for the treatment of HER2-positive breast cancer. HL02 is a biosimilar of trastuzumab, which targets the extracellular domain IV of the Human Epidermal Growth Factor Receptor 2 (HER2), inhibiting ligand-independent HER2 signaling and mediating antibody-dependent cellular cytotoxicity (ADCC). WBP257 is a biosimilar of pertuzumab, which binds to domain II of HER2, specifically inhibiting the dimerization of HER2 with other HER family receptors, such as HER3. By binding to different epitopes on the HER2 receptor, the combination provides a more comprehensive blockade of HER2-mediated signaling pathways (including PI3K/Akt and MAPK), leading to enhanced inhibition of tumor cell growth and survival compared to monotherapy. This combination is primarily being evaluated in Phase 3 clinical trials in China as a neoadjuvant treatment for HER2-positive breast cancer.
