This is a combination regimen of three HER2-targeted therapies used primarily in the treatment of HER2-positive breast cancer. Trastuzumab and pertuzumab are monoclonal antibodies that bind to different epitopes on the human epidermal growth factor receptor 2 (HER2), inhibiting its signaling and promoting antibody-dependent cellular cytotoxicity. Trastuzumab emtansine (T-DM1) is an antibody-drug conjugate that combines trastuzumab with DM1, a cytotoxic agent that inhibits microtubule polymerization, thereby inducing cell death specifically in HER2-overexpressing cells. This combination has been studied for synergistic activity and improved outcomes in advanced or metastatic HER2-positive breast cancer[1][3][6].
