trastuzumab + pyrotinib + dalpiciclib + endocrine therapy is a combination therapeutic regimen being investigated for the treatment of advanced hormone receptor-positive (HR+) and human epidermal growth factor receptor 2-positive (HER2+) breast cancer. This regimen combines multiple targeted approaches: pyrotinib, an oral, irreversible pan-ErbB receptor tyrosine kinase inhibitor targeting EGFR, HER2, and HER4; trastuzumab, a monoclonal antibody targeting the extracellular domain of HER2; dalpiciclib (SHR6390), a selective CDK4/6 inhibitor that blocks cell cycle progression; and endocrine therapy to inhibit estrogen-driven tumor growth. The combination is currently being evaluated in a Phase II clinical trial led by Peking University Cancer Hospital & Institute to improve survival outcomes in patients with HER2-positive advanced breast cancer. Pyrotinib and dalpiciclib are both developed by Jiangsu Hengrui Pharmaceutical, while trastuzumab is a standard-of-care HER2-targeted biologic.
