Overview

Trastuzumab ADC 1 refers to an antibody-drug conjugate (ADC) that utilizes the HER2-targeted monoclonal antibody trastuzumab (or a biosimilar/derivative thereof) conjugated to a cytotoxic payload (such as a microtubule inhibitor like DM1 or MMAE, or a topoisomerase I inhibitor like DXd) via a chemical linker. Trastuzumab ADCs are designed to selectively deliver highly potent cytotoxic agents to HER2-expressing cancer cells, minimizing systemic toxicity. Upon binding to HER2 on the cell surface, the ADC is internalized via receptor-mediated endocytosis, and the payload is released intracellularly to induce cell death. Examples of approved trastuzumab ADCs include ado-trastuzumab emtansine (T-DM1) and fam-trastuzumab deruxtecan (T-DXd).
