Trastuzumab and hyaluronidase-oysk is a fixed-dose combination of two agents for subcutaneous injection. It contains trastuzumab, a recombinant humanized monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2; also known as receptor tyrosine-protein kinase erbB-2), and hyaluronidase-oysk, a recombinant human hyaluronidase PH20 enzyme. Trastuzumab binds to HER2 on cancer cells, blocking their ability to send growth signals and mediating antibody-dependent cellular cytotoxicity (ADCC) against HER2-overexpressing tumor cells. Hyaluronidase temporarily breaks down the extracellular matrix component hyaluronan at the injection site, increasing tissue permeability and facilitating rapid absorption of trastuzumab into systemic circulation. This formulation allows for faster subcutaneous administration compared to intravenous trastuzumab. The drug is approved for use in adults with HER2-positive breast cancer—both early-stage (as adjuvant therapy) and metastatic settings—either in combination with other chemotherapies or as monotherapy after prior treatments[1][3][4][6].
