Overview

Trastuzumab-dkst is a biosimilar to trastuzumab, marketed under the brand name Ogivri. It is a humanized IgG1 kappa monoclonal antibody that targets the human epidermal growth factor receptor 2 (HER2). Trastuzumab-dkst binds to an epitope on the extracellular juxtamembrane domain of HER2, inhibiting its activity and leading to downregulation of downstream survival signaling pathways, cell cycle arrest, and apoptosis. The drug also activates immune-mediated mechanisms such as antibody-dependent cellular cytotoxicity (ADCC). Trastuzumab-dkst is indicated for the treatment of HER2-overexpressing breast cancer (early-stage and metastatic) and HER2-overexpressing metastatic gastric or gastroesophageal junction cancer. It should only be used in patients whose tumors have demonstrated HER2 overexpression[1][3][4][6].
