Overview

Trastuzumab duocarmazine (SYD985) is an investigational next-generation antibody-drug conjugate (ADC) developed by Byondis for the treatment of HER2-positive cancers, primarily metastatic breast cancer and endometrial cancer. It consists of the anti-HER2 monoclonal antibody trastuzumab linked to a cytotoxic agent via a cleavable linker. The antibody binds to HER2 on tumor cells, is internalized, and releases the cytotoxin after proteolytic cleavage, causing DNA damage and cell death. BYON5667 is another investigational agent from Byondis; however, detailed public information about its mechanism or clinical status is limited as of April 2025. The combination represents an experimental therapy under clinical investigation for solid tumors overexpressing HER2[1][3][4][5].
