trastuzumab-NMS-P945 is a preclinical-stage antibody-drug conjugate (ADC) developed by Nerviano Medical Sciences. It consists of the anti-HER2 monoclonal antibody trastuzumab conjugated to NMS-P945, a novel protease-cleavable thienoduocarmycin-like payload-linker containing the potent DNA minor groove alkylating agent NMS-P528. Upon binding to HER2 on cancer cells, the ADC is internalized and lysosomally degraded, releasing the active payload which induces DNA damage, cell cycle arrest, and apoptosis. In preclinical models of HER2-positive breast cancer, trastuzumab-NMS-P945 has demonstrated robust antitumor efficacy, bystander killing effects, and immunogenic cell death.
