Trastuzumab rezetecan + adebrelimab is a combination therapy consisting of a HER2-targeted antibody-drug conjugate (ADC) and a PD-L1 immune checkpoint inhibitor. Trastuzumab rezetecan (SHR-A1811) is composed of a humanized anti-HER2 monoclonal antibody conjugated to a cytotoxic topoisomerase I inhibitor payload via a cleavable linker. It is designed to deliver potent cytotoxicity directly to HER2-expressing tumor cells. Adebrelimab (SHR-1316) is a humanized monoclonal antibody that targets Programmed cell death 1 ligand 1 (PD-L1), blocking its interaction with PD-1 and B7.1 receptors to restore and enhance anti-tumor T-cell activity. This combination is being evaluated in Phase II clinical trials for the treatment of HER2-positive or HER2-expressing solid tumors, including muscle-invasive bladder cancer (MIBC) and urothelial carcinoma, aiming to leverage the synergistic effects of direct tumor cell killing and immune system activation.
