Trazodone hydrochloride (new polymer formulation) is an extended-release, small molecule antidepressant being developed by Aziende Chimiche Riunite Angelini Francesco S.p.A (Angelini Group) for the treatment of major depressive disorder. This specific product utilizes a novel polymer-based controlled-release technology designed to optimize the pharmacokinetic profile of trazodone, distinguishing it from previous extended-release versions such as the Contramid-based formulation (marketed as Oleptro or Trittico). Trazodone acts as a serotonin antagonist and reuptake inhibitor (SARI), primarily functioning as an antagonist at the 5-HT2A and 5-HT2C receptors and as an inhibitor of the serotonin transporter (SERT). It also exhibits antagonistic activity at alpha-1 adrenergic and histamine H1 receptors, which contributes to its clinical profile. The new polymer formulation is currently undergoing Phase 1 clinical evaluation to establish its bioavailability and bioequivalence relative to existing once-a-day trazodone products.
