Overview

Trazpiroben (TAK-906) is a peripherally selective dopamine D2 and D3 receptor antagonist developed for the treatment of gastroparesis. It was designed to minimize central nervous system and cardiovascular side effects commonly associated with other dopamine antagonists by exhibiting minimal brain penetration and low affinity for the hERG potassium channel. Trazpiroben increases prolactin levels in humans as a marker of peripheral D2 receptor antagonism. The drug was originally developed by Altos Therapeutics and later by Takeda Oncology but its clinical development was discontinued before April 2022[1][4][5][6].
