Overview

TRBC2 CAR-T is an investigational autologous chimeric antigen receptor T cell therapy engineered to target the T cell receptor beta-chain constant region 2 (TRBC2), which is expressed on a mutually exclusive subset of TCRαβ T cells. The therapy uses a TRBC2-specific binder (such as the KFN antibody) in the CAR construct to enable selective recognition and elimination of malignant T cells that clonally express TRBC2, while sparing normal TRBC1-expressing T cells and thereby preserving nonmalignant T cell immunity. This approach aims to treat TRBC2-positive peripheral T cell malignancies, addressing the challenge of safely targeting T cell cancers without causing profound immunosuppression or fratricide among T cells. The therapy is in preclinical and early clinical development stages, complementing similar advances for anti-TRBC1 CAR-T cell therapies[1][3][6][9].
