This regimen combines: - TRC105 (carotuximab), a chimeric IgG1 monoclonal antibody against CD105 (endoglin), which targets proliferating endothelial cells to inhibit angiogenesis, often by inducing apoptosis in CD105-positive cells and potentially counteracting resistance mechanisms to anti-VEGF therapy[1][3][5]. - Bevacizumab, a monoclonal antibody against vascular endothelial growth factor A (VEGF-A), inhibiting tumor angiogenesis[3]. - Paclitaxel, a microtubule-stabilizing small molecule cytotoxic chemotherapy that inhibits cell division[2][4][6]. - Carboplatin, a platinum-based cytotoxic chemotherapeutic, inducing DNA crosslinking and apoptosis[2][4][6]. The rationale for the combination is twofold: dual inhibition of tumor vasculature (TRC105/CD105 and bevacizumab/VEGF), and cytotoxic reduction of tumor burden (paclitaxel and carboplatin).
