Overview

TRC694 is an orally bioavailable, small-molecule inhibitor of NF-κB-inducing kinase (NIK), developed initially by Janssen Pharmaceutica and later licensed to TRACON Pharmaceuticals for oncology indications. By selectively inhibiting NIK, TRC694 blocks processing of p100 to p52 and thereby suppresses the alternative NF-κB signaling pathway, leading to reduced survival and proliferation of tumor cells with constitutive NIK activation, such as multiple myeloma models harboring TRAF3 mutations. Preclinical studies demonstrated that TRC694 potently inhibited p100-to-p52 processing in multiple myeloma cell lines, strongly suppressed tumor growth in RPMI-8226 and MM.1S xenograft models, and represented the first NIK inhibitor to show antitumor efficacy in mouse tumor models, supporting its evaluation in hematologic malignancies and other NF-κB–driven cancers.[1][4][7][9][11]
