trCAR27-28.3Z NK cells are an experimental chimeric antigen receptor (CAR) natural killer (NK) cell therapy designed to target CD70-expressing malignancies. Developed by researchers at MD Anderson Cancer Center, the construct features a truncated CD27 receptor (the natural ligand for CD70) as the antigen-binding domain, fused to CD28 costimulatory and CD3-zeta (3Z) signaling domains. CD70 is a pan-cancer antigen expressed in various hematologic cancers like acute myeloid leukemia (AML) and lymphoma, as well as solid tumors such as renal cell carcinoma, lung cancer, and melanoma, while remaining largely absent from normal tissues. This CAR-NK approach leverages the intrinsic anti-tumor properties of NK cells and their potential for allogeneic use with minimal risk of graft-versus-host disease (GvHD).
