Trebananib + liposomal doxorubicin is a combination therapy investigated primarily for the treatment of recurrent epithelial ovarian cancer. Trebananib (AMG 386) is a recombinant peptide-Fc fusion protein that inhibits angiogenesis by blocking the interaction of angiopoietin 1 and 2 with the Tie2 receptor, thereby disrupting blood vessel formation essential for tumor growth[4][5]. Liposomal doxorubicin is an anthracycline chemotherapy agent encapsulated in pegylated liposomes to prolong circulation time and enhance delivery to tumors. Its mechanism involves intercalation into DNA, inhibition of topoisomerase II, and generation of free radicals leading to cell death[6][7][8]. The combination has been studied in phase III clinical trials for recurrent ovarian cancer but did not show significant improvement in progression-free survival compared to monotherapy with pegylated liposomal doxorubicin[4][5].
