TREG-AI-1 is a preclinical-stage tolerogenic immunotherapy candidate being developed by TregTherapeutics for the treatment of systemic autoimmune diseases (specifically designated as Systemic Autoimmune 1). It is built on the company's patented TILAC (Tolerance Induction via Linked Adjuvant/Cytokine conjugate) platform, which was originally developed at East Carolina University. The TILAC platform is designed to restore immune tolerance by shifting the dendritic cell environment from immunogenic to tolerogenic signaling. This tolerogenic environment promotes the expansion of regulatory T cells (Tregs) to suppress disease-associated inflammation without causing broad immune suppression. TREG-AI-1 utilizes an alum-based adjuvant conjugated to an anti-inflammatory cytokine to induce antigen-agnostic immune tolerance.
