TREG-AI-2 is a preclinical-stage tolerogenic immunotherapy being developed by TregTherapeutics (exclusively licensed from East Carolina University) for the treatment of systemic autoimmune diseases (specifically designated as Systemic Autoimmune Disease 2). Developed using the proprietary TILAC (Tolerance Induction via Linked Adjuvant/Cytokine conjugate) platform, TREG-AI-2 consists of an alum-based adjuvant conjugated to an anti-inflammatory cytokine. The therapy is designed to shift the dendritic cell microenvironment from immunogenic to tolerogenic signaling, promoting the expansion of antigen-specific regulatory T cells (Tregs) to restore immune tolerance and suppress pathogenic inflammation without causing systemic immunosuppression.
