Overview

Treg and Tcon Immunotherapy is an investigational cellular immunotherapy developed by Stanford University for the treatment of advanced hematologic malignancies in patients undergoing allogeneic hematopoietic cell transplantation (HCT). The therapy involves the sequential infusion of donor-derived, freshly isolated CD4+CD25+FOXP3+ regulatory T cells (Tregs) and conventional T cells (Tcons). The Tregs are administered four days prior to the Tcons and CD34+ hematopoietic stem cells to suppress the initiation of graft-versus-host disease (GVHD). The subsequent Tcon infusion is intended to facilitate immune reconstitution and maintain the graft-versus-leukemia (GVL) effect. This approach is designed to eliminate the need for standard post-transplant pharmacological GVHD prophylaxis, thereby potentially reducing transplant-related morbidity and improving long-term outcomes for patients with conditions such as acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), and chronic myeloid leukemia (CML).
