Trelagliptin is a highly selective, long‐acting, orally active dipeptidyl peptidase‐4 (DPP‐4) inhibitor developed for the treatment of type 2 diabetes mellitus. It is notable as the first once-weekly oral DPP‐4 inhibitor, offering an alternative to daily dosing regimens. Trelagliptin works by inhibiting DPP‐4, an enzyme that degrades incretin hormones such as glucagon-like peptide 1 (GLP‑1) and glucose-dependent insulinotropic polypeptide (GIP), thereby prolonging their activity and improving glycemic control. Clinical studies have demonstrated its efficacy in reducing hemoglobin A1c (HbA1c) with a safety profile comparable to other DPP‑4 inhibitors. Trelagliptin was developed by Takeda and approved in Japan and India for type 2 diabetes; development was discontinued in the US and EU due to regulatory cost considerations[2][4][5][8].
