Overview
Trelarafen (ZR-01) is a small molecule BRAF kinase inhibitor being developed by Nanjing Zhongrui Pharmaceutical for the treatment of acute ischemic stroke (AIS). While BRAF inhibitors are traditionally utilized in oncology, Trelarafen is being investigated for its neuroprotective potential. The drug targets the BRAF/MEK/ERK signaling pathway, which is a key mediator of neuroinflammation, blood-brain barrier disruption, and neuronal apoptosis following an ischemic event. By inhibiting this pathway, Trelarafen aims to reduce brain tissue damage and improve functional recovery in stroke patients. It is currently being evaluated in Phase 1 clinical trials as a continuous intravenous infusion to establish its safety, tolerability, and pharmacokinetic profile in humans.
