TREM2 lentivirus is a gene therapy platform designed to deliver and overexpress the Triggering Receptor Expressed on Myeloid cells 2 (TREM2) gene. TREM2 is a transmembrane receptor primarily expressed on microglia in the central nervous system and macrophages in the periphery, where it plays a critical role in sensing lipid ligands, amyloid-beta, and apoptotic neurons. Mutations in TREM2 are strongly associated with an increased risk of Alzheimer's disease due to impaired microglial phagocytosis and altered neuroinflammatory responses. By utilizing a lentiviral vector, this approach enables stable, long-term expression of the TREM2 protein in target cells. In preclinical models of neurodegeneration, TREM2 lentivirus is used to enhance microglial clearance of pathological protein aggregates, promote cell survival, and modulate the inflammatory environment to slow disease progression. While widely utilized as a research-use-only (RUO) tool, it represents a potential therapeutic strategy for Alzheimer's disease and other tauopathies.
