Overview

Tremelimumab is a fully human IgG2 monoclonal antibody immune checkpoint inhibitor that targets cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), a negative regulator of T-cell activation. By binding to CTLA-4 and blocking its interaction with ligands CD80 and CD86, tremelimumab enhances T-cell activation and proliferation, resulting in increased antitumor immune activity. It is primarily used in combination with durvalumab for the treatment of unresectable hepatocellular carcinoma (HCC) and metastatic non-small cell lung cancer (NSCLC) without EGFR or ALK mutations. Tremelimumab was developed as an immunotherapy for cancer by AstraZeneca/MedImmune.
