Trenbolone is a synthetic androgen and anabolic steroid (AAS) of the nandrolone group that was developed for veterinary use to promote muscle growth and appetite in livestock[1][2][4]. It is not approved for human use but has gained popularity among bodybuilders and athletes as a performance-enhancing drug due to its potent anabolic effects[7]. Trenbolone acts primarily as an agonist of the androgen receptor (AR), with approximately three times higher affinity than testosterone[3]. This leads to increased protein synthesis, nitrogen retention in muscles, stimulation of appetite, decreased catabolism during administration, and significant muscle hypertrophy[2][3][7]. Trenbolone also binds with high affinity to the progesterone receptor and glucocorticoid receptor[2], contributing to its complex hormonal effects. It is typically administered as ester prodrugs such as trenbolone acetate or enanthate; these are hydrolyzed in vivo to release active trenbolone[1][5]. The drug increases levels of insulin-like growth factor-1 (IGF-1), further promoting muscle recovery and growth[5]. Its mechanism involves both genomic pathways via AR activation—leading to upregulation of proanabolic genes—and non-genomic modulation affecting immune cells and other tissues[3][6].
