Treo is an over-the-counter effervescent analgesic brand containing a fixed-dose combination of acetylsalicylic acid and caffeine, marketed primarily in Nordic and European countries for short-term relief of mild to moderate pain such as headache, migraine, toothache, dysmenorrhea, and fever. The pharmacology is driven by acetylsalicylic acid, a small-molecule nonsteroidal anti-inflammatory drug (NSAID) that irreversibly inhibits cyclooxygenase (COX-1 and COX-2), reducing prostaglandin synthesis to produce analgesic, antipyretic, and anti-inflammatory effects. Caffeine, a methylxanthine CNS stimulant, enhances analgesia and can constrict dilated cerebral blood vessels, which is particularly relevant in migraine, while also counteracting sedation and improving perceived alertness. Treo is formulated as effervescent tablets that are dissolved in water before administration, resulting in rapid onset of action and somewhat improved gastric tolerability compared with solid acetylsalicylic acid tablets.
