HF1K16 (tretinoin liposome) is an investigational pegylated liposome formulation of all-trans retinoic acid (ATRA, also known as tretinoin) developed by HighField Biopharmaceuticals. Unlike traditional applications of ATRA in acute promyelocytic leukemia (APL), HF1K16 is designed to target the immunosuppressive tumor microenvironment in solid tumors. It acts by infiltrating the tumor microenvironment and releasing ATRA, which binds to retinoic acid receptors (RARs) and promotes the differentiation and maturation of myeloid-derived suppressor cells (MDSCs) into functional antigen-presenting cells like dendritic cells. This depletion of MDSCs restores T-cell activity and remodels the tumor microenvironment from immunosuppressive to immunoreactive. HF1K16 is being evaluated in clinical trials for the treatment of advanced solid tumors, including recurrent glioma, pancreatic cancer, and non-small cell lung cancer.
