Drug intelligence / Profile preview
Tretinoin microsphere is a topical formulation of the retinoid drug tretinoin, designed for the treatment of acne vulgaris. It utilizes patented porous polymeric microspheres (Microsponges or similar acrylates copolymer systems) to encapsulate and gradually release tretinoin into the skin. This controlled-release delivery improves tolerability by reducing irritation compared to conventional formulations and enhances photostability. Tretinoin acts primarily by decreasing cohesiveness of follicular epithelial cells, reducing microcomedone formation, stimulating mitotic activity, and increasing turnover of follicular epithelial cells—leading to extrusion of comedones. Mechanistically, it activates retinoic acid receptors (RARα, RARβ, RARγ), modifying gene expression related to cell growth and differentiation; however, its exact clinical mechanism remains not fully established[1][2][6][9].
