TRI-1144 is a second-generation HIV-1 fusion inhibitor peptide consisting of 38 amino acids. Developed by Trimeris, it is designed to block the entry of HIV-1 into host cells by targeting the viral fusion machinery. The peptide is an optimized version of earlier fusion inhibitors, such as enfuvirtide, intended to provide increased potency and a higher genetic barrier to resistance. Research has explored the use of half-life extension technologies, specifically PEGylation using β-eliminative linkers, to improve its pharmacokinetic profile and enable less frequent dosing (e.g., weekly administration).
