Overview

TRI-611 is a preclinical, orally bioavailable small-molecule molecular glue degrader developed by Triana Biomedicines to selectively degrade anaplastic lymphoma kinase (ALK) fusion oncoproteins, including variants resistant to current tyrosine kinase inhibitors, in anaplastic lymphoma kinase–positive (ALK+) non-small cell lung cancer (NSCLC). It is described as a selective, CNS‑penetrant ALK-targeted degrader with strong antiproliferative and anti-tumor activity in preclinical ALK-fusion–positive and ALK TKI–refractory NSCLC models, with broad selectivity across the proteome that minimizes off‑target kinase effects such as NTRK1 while maintaining sustained target coverage, including in the central nervous system.[1][2][11][14] TRI-611 is currently in IND-enabling development with Series B financing dedicated to advancing it to clinical proof of concept in ALK+ NSCLC, positioning it as a potential first-in-class clinical molecular glue degrader of an oncogenic fusion protein and a next-generation option for patients who develop resistance or tolerability issues on existing ALK tyrosine kinase inhibitors.[1][2][7][8][10][13]
