Overview

Tri-Ad5 is an investigational **three-component therapeutic cancer-vaccine regimen** comprising replication-defective recombinant human adenovirus serotype 5 vectors encoding carcinoembryonic antigen-related cell adhesion molecule 5, mucin-1, and brachyury. The three component vaccines are ETBX-011, ETBX-061, and ETBX-051, respectively. Following subcutaneous administration, antigen expression is intended to prime antigen-specific cytotoxic T-cell responses against tumor or premalignant cells expressing these tumor-associated antigens. The vector backbone has deletions in adenoviral E1, E2b, and E3 genes. Tri-Ad5 is being developed by ImmunityBio and is in Phase 2 development with nogapendekin alfa inbakicept for cancer prevention in Lynch syndrome; it is also being studied in combination regimens for metastatic colorectal cancer.
