Overview

TriAb is an anti-idiotypic monoclonal antibody, specifically designated as 6D11, that was developed as a therapeutic cancer vaccine to target the Lewis Y (LeY) carbohydrate antigen. Lewis Y is a difucosylated oligosaccharide that is highly expressed on the surface of various epithelial cancers, including breast, ovarian, colorectal, and small cell lung cancers, while having limited expression on normal tissues. As an anti-idiotypic antibody (Ab2), TriAb mimics the three-dimensional structure of the LeY antigen, thereby acting as a surrogate to stimulate the patient's immune system to produce anti-anti-idiotypic antibodies (Ab3). These induced antibodies are specific for the original LeY antigen and can mediate tumor cell killing through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC). Developed by Titan Pharmaceuticals, TriAb reached Phase 3 clinical trials for breast cancer but was ultimately discontinued after failing to meet primary efficacy endpoints.
