Overview

TriAd is a trivalent therapeutic cancer vaccine cocktail consisting of three distinct second-generation (E1-, E2b-deleted) adenovirus type 5 (Ad5) vectors, each encoding a different tumor-associated antigen: Carcinoembryonic Antigen (CEA; ETBX-011), Mucin 1 (MUC1; ETBX-051), and Brachyury (ETBX-061). Developed by ImmunityBio, the vaccine platform is designed to overcome pre-existing Ad5 immunity and induce robust Th1-mediated T-cell responses against multiple targets simultaneously. It is frequently evaluated in combination with other immunotherapies, such as the IL-15 superagonist N-803 (Anktiva) and the PD-L1/TGF-beta trap bintrafusp alfa (M7824), for the treatment of various solid tumors, including head and neck squamous cell carcinoma, colorectal cancer, and chordoma.
