Overview

TriAdeno Vaccine is an investigational therapeutic cancer vaccine developed by the National Cancer Institute (NCI). It utilizes a viral vector platform consisting of three separate adenovirus type 5 (Ad5) vectors, each engineered to express a specific tumor-associated antigen: Carcinoembryonic Antigen (CEA), Brachyury, and Mucin 1 (MUC1). These antigens are frequently overexpressed in various solid tumors, including colorectal cancer. The vaccine is designed to stimulate the patient's immune system, specifically inducing a T-cell response against tumor cells harboring these proteins. In clinical settings, such as the STAR15 trial, it is being evaluated as part of a multi-agent immunotherapy regimen (including retifanlimab, N-803, and SX-682) for the treatment of metastatic colorectal cancer (mCRC).
