Drug intelligence / Profile preview
FX006 (Zilretta) is an extended-release, intra-articular formulation of triamcinolone acetonide, a synthetic corticosteroid. Developed using a proprietary poly(lactic-co-glycolic acid) (PLGA) microsphere technology, it is designed to provide sustained pain relief by maintaining therapeutic concentrations of the drug within the joint space for an extended period. Triamcinolone acetonide acts as a potent agonist of the glucocorticoid receptor, leading to the suppression of inflammatory cytokines and mediators. While Zilretta is FDA-approved for the management of osteoarthritis pain of the knee, FX006 has been evaluated in Phase 2 clinical trials (such as the RANGE study) for other indications, including glenohumeral osteoarthritis and adhesive capsulitis (frozen shoulder). The formulation aims to overcome the rapid clearance typically seen with immediate-release corticosteroid injections.
