Overview

This investigational combination therapy, developed and studied by the University of Washington, consists of the antimetabolite 5-fluorouracil (5-FU) and the corticosteroid triamcinolone acetonide. It is specifically designed for the treatment of chalazia (eyelid cysts) and is administered via local intralesional injection directly into the lesion. The combination typically utilizes a 9:1 ratio of 5-FU to triamcinolone acetonide. 5-FU acts as a chemotherapy agent that inhibits thymidylate synthase, thereby disrupting DNA synthesis in the proliferating cells of the chalazion, while triamcinolone acetonide provides potent anti-inflammatory effects. This dual mechanism aims to reduce the size of the chalazion and improve cosmetic appearance while minimizing the side effects associated with high-dose steroid injections, such as skin hypopigmentation and increased intraocular pressure.
