TRIB3 siRNA is a small interfering RNA (siRNA) designed to silence the expression of tribbles pseudokinase 3 (TRIB3), a member of the tribbles family of scaffold proteins. TRIB3 lacks significant kinase activity but modulates various signaling pathways, including the AKT, MAPK, and TGF-β pathways, and is often upregulated in response to cellular stress. In oncological research, particularly in endometrial cancer, TRIB3 has been identified as a key regulator of cancer stem cell (CSC) maintenance through its interaction with the transcription factor ELF4, which subsequently promotes the expression of CDK6. By targeting TRIB3 mRNA for degradation, TRIB3 siRNA reduces TRIB3 protein levels, leading to decreased CDK6 expression and inhibition of CSC-related properties such as tumorsphere formation and the expression of stemness markers like OCT4 and NANOG.
