Overview

Tribendimidine is a broad-spectrum anthelmintic agent developed through a long-term research collaboration between the National Institute of Parasitic Diseases (NIPD) in China and the Swiss Tropical and Public Health Institute (Swiss TPH). A symmetric derivative of amidantel, it is particularly effective against soil-transmitted helminths such as hookworms (*Necator americanus* and *Ancylostoma duodenale*) and foodborne trematodes, including the liver flukes *Clonorchis sinensis* and *Opisthorchis viverrini*. Its mechanism of action involves acting as a cholinergic agonist at nicotinic acetylcholine receptors (nAChR) on the body wall muscles of parasites, inducing spastic paralysis. The drug is rapidly metabolized into its active metabolite, deacetylated amidantel (dADT). Registered in China since 2004, it is being evaluated as a high-efficacy alternative to praziquantel and albendazole, especially in combination regimens to overcome drug resistance in neglected tropical diseases.
