Tribenoside is a small molecule vasoprotective and anti-inflammatory drug primarily used for the treatment of hemorrhoids and venous circulation disorders such as varicose veins and phlebitis[1][2][3][4]. It exhibits mild anti-inflammatory, analgesic, wound-healing, antitoxic, fibrinolysis-promoting, membrane-stabilizing, and venotropic properties[6][8]. Tribenoside acts by reducing vascular permeability and causing vasoconstriction. Mechanistically, it antagonizes inflammatory mediators including histamine, serotonin (5-HT), bradykinin and others[3]. It also stimulates laminin α5 production and laminin-332 deposition to support basement membrane repair during wound healing[1]. Unlike corticosteroids or NSAIDs it does not adversely affect the gastrointestinal tract or immune system[6][8]. Tribenoside is often formulated in combination with lidocaine for topical use in hemorrhoid management under brand names such as Procto-Glyvenol[5][8].
