Overview

Triclabendazole is a benzimidazole-derivative anthelmintic drug used to treat fascioliasis, a parasitic infection caused by liver flukes (Fasciola hepatica and Fasciola gigantica) in adults and children aged 6 years and older. It is the only FDA-approved treatment for human fascioliasis. Triclabendazole works by binding to β-tubulin in the parasite, inhibiting microtubule formation, which disrupts essential cellular processes such as protein and enzyme synthesis, motility, and egg production. Its active metabolites (sulfoxide and sulfone) are absorbed by the worm's tegument (outer surface), leading to metabolic disturbances that kill both immature and mature worms. The precise mechanism of action is not fully elucidated but involves inhibition of tubulin function specific to Fasciola species[1][3][5][6].
